Pancreatic Cancer Cell Line Establishment and Characterization

Pancreatic Cancer Cell Line Establishment and Characterization

Alfa Cytology is a leading provider of pancreatic cancer (PC) drug development services. We provide a range of services around PC, including important parts such as basic research, drug development, and diagnosis of PC. We are committed to providing our customers with optimal customized services to support basic and translational PC research projects. Here, we offer PC cell line establishment and characterization services to help researchers and professionals to confirm therapeutic efficacy or identify additional options.

More biological resources are needed to study PC

Malignant cells indicating adenocarcinoma.

Although relatively uncommon worldwide (2.5% of all forms of cancers), its poor prognosis ranks PC the fourth or fifth in malignancy mortalities. Surgery is still the main treatment for PC, but only 15% to 20% of PC patients can be resected at diagnosis with a poor prognosis after resection. Although the currently available adjuvant therapies appear to provide a survival benefit, they are only moderate and standard protocols have not been established. On the other hand, PC stimulates a pro-fibroproliferative response in vivo, thereby expanding the space between tumor cells with reactive non-tumor components (e.g., inflammatory cells and fibroblasts), and reducing the number of viable cancer cells available in each macroscopic field of view. Based on this biological nature, only 53 PC cell lines are currently available in this research area. Not to mention, some available cell lines are already outdated. Therefore, there is an urgent need to establish and develop more biological resources to facilitate the study of PC.

The service offering at Alfa Cytology

  • Establishment of cell lines and cell culture

The derivation and short-term culture of solid tumor primary cells are of great importance in individualized cancer therapy. Newly established pancreatic cancer cell lines may provide sufficient models for studying the broader biological and molecular features of PC. We have established cell culture-based methods for tumor cell isolation and expansion for further molecular and functional testing, including in vitro 2D and 3D culture systems.

  • Cell growth properties and general characterization

Molecular characterization of cultured cancer cells is performed by various methods and functional assays including cell viability and drug sensitivity assays.

-Cell growth rate in established PC cell lines
-Cell viability in established PC cell lines
-Assessment of drug sensitivity in established PC cell lines

  • Whole exome sequencing (WES) analysis

WES analysis is applied to investigate the mutational context of the established PC cell lines. And further analysis of mutations, such as gene set enrichment analysis is performed, to find representative pathways that are aberrated in established PC cell lines.

-Variant classification
-Identification of the mutation patterns of PC cell lines
-Gene set enrichment analysis
-Targeted gene prediction

  • Reverse transcription PCR and expression of molecular markers
Scientists work with cell cultures under the sterile hood.
