Ca2+ imaging is widely used in the fields of neurobiology, cell biology, physiology, developmental biology, and pharmacology. Alfa Cytology uses indicators that can sense Ca2+ concentration to convert changes in calcium concentration into fluorescent signals, and the displayed spectra can be detected by a variety of methods such as fluorescence microscopy, flow cytometry, fluorescence spectroscopy, and fluorescence enzyme markers. Intracellular Ca2+ is involved in the facilitation of inter-synaptic signaling in neurons, and the level of intracellular Ca2+ concentration detected by calcium imaging can reflect the function of neuron-like cells. Two types of Ca2+ indicators, chemical indicators, and genetically encoded indicators are now commonly used, such as GCaMP. GCaMP consists of circularly permuted green fluorescent proteins (cpGFP), calmodulin (CaM), and a peptide sequence M13 of myosin light chain kinase. When Ca2+ binds CaM, CaM conformationally changes and its hinge region binds M13, causing cpGFP to fluoresce intensely with increased protonation of its chromophore and an increase in absorbance in the presence of light at specific wavelengths. 300s after activation of the indicator, the fluorescent signal filled the whole cell, including dendrites and dendritic spines.
