Malignant gliomas account for 70% of primary malignant brain tumors, with an annual incidence of 5/100,000 and more than 14,000 new cases each year. Researchers have identified a specific subpopulation of cells in brain tumors. This subpopulation of cells has the ability of self-renewal, proliferation, and multi-differentiation potential, which is brain tumor stem cells (BTSCs). And the presence of BTSCs may be the root cause of brain tumorigenesis, recurrence, and drug resistance.

BTSCs analysis services that we offer

Neurospheres are composed of stem cells, progenitor cells, and terminally differentiated cells. During the continuous passages, differentiated cells die quickly, and stem cells proliferate and this process can be repeated. This results in a large, stable population of neural stem cells in the culture system. With this technology, Alfa Cytology can isolate human brain tumor specimens. We apply the direct culture method to obtain BTSCs, identify them, and analyze their biological properties. This can provide you with reliable information for further brain tumor research.

General analysis flow and content

  • Isolation and culture of brain tumor cells. Human brain tumor tissue is taken, cells are isolated by enzymatic digestion, and inoculated in a culture medium for culture.
  • Analysis of neurosphere formation rate. Brain tumor cells were inoculated in 96-well plates, one cell per well. The wells containing individual cells were observed and labeled under the microscope. The growth and proliferation of cells were observed and the neurosphere formation rate was counted after some time.
  • Detection of surface antigens on neurospheres neural stem cells. We collected neurospheres and performed staining, primary antibody, and DAPI staining to seal the slides. They were observed under a fluorescence microscope and photographed.
  • Neurosphere multi-differentiation potential assay. Neurosphere cells are inoculated in a differentiation solution at an appropriate density for some time, and primary antibody. DAPI staining is blocked, observed under a fluorescence microscope, and photographed.
