Transmonosynaptic Retrograde and Biological Markers Neural Stem Cell Lines

The vesicular membrane glycoprotein (G) of RV is a protein essential for its retrograde trans-synaptic movement. Its receptor is abundantly distributed at axon terminals and can retrograde along the axon into the neuronal cytosol to initiate viral replication after infection. G protein-deficient RV (RV-∆G) loses its trans-synaptic ability, while its replication and transcription are unaffected (sustained high abundance of exogenous genes can be expressed), and it can retrograde and highly label neuronal fine morphology. Compensation of the RV-G protein, in turn, assists RV-∆G in retrograde trans-synaptic infection of higher-level neurons, thus enabling labeling across monosynaptic neural networks.

Alfa Cytology uses recombinant A-avian sarcoma-leukosis virus (ASLV) outer membrane protein (EnvA) fusion proteins to package viral particles formed by RV, which specifically recognizes EnvA's receptor TVA to mediate viral infection of cells specifically. We used Cre transgenic mice in combination with Cre-LoxP to control the expression of TVA and G proteins in AAV helper viruses, which enables the expression of TVA and G proteins only in specific types of neurons in specific regions. Thus, RV-En-vA-∆G was used to achieve reverse trans-single-stage synaptic labeling of specific types of neurons.

RV-En-vA-∆G-dsRed binds to G protein-expressing helper viruses to achieve neuronal labeling across single synapses. Red fluorescent protein-labeled neurons were seen in brain regions such as the hippocampus on both sides, the dorsolateral thalamic nucleus, the anterior dorsal thalamic nucleus, and the paraventricular nucleus of the thalamus.
