Antigen-Specific T Cell Response Assay Services

Test whether candidate antigens or vaccine-related samples elicit a measurable, specific T cell response using an assay configuration matched to your research question.

Functional Testing Beyond Antigen Prediction

A predicted epitope is a starting point, but it does not establish that T cells recognize the antigen. Alfa Cytology offers antigen-specific T cell response assay services to help cancer vaccine researchers compare candidate antigens, examine vaccine-associated immune responses, and prioritize follow-up experiments. The study is built around the cells and antigen-recognition system available for your project.

A single cytokine measurement rarely answers every development question. Some projects need a sensitive screen for responding cells; others require phenotype, proliferation, or a secondary functional readout. We define whether the experiment measures a direct ex vivo response, a response after in vitro expansion, or a newly induced response before selecting methods. These results are reported separately rather than combined into one apparent response rate.

Assay Options and What They Measure

ELISpot-Based Response Detection

Consider IFN-γ ELISpot to quantify cytokine-secreting cells after antigen stimulation. The plan specifies cells per well, controls, background handling, and response criteria. Additional cytokine formats or method-development work are scoped separately.

Activation and Immune Phenotyping

Use a project-selected flow cytometry panel to identify responding populations and activation-associated changes. Marker combinations and timing are selected for CD4 or CD8 questions; marker positivity alone is not treated as proof of tumor-cell killing.

Antigen-Driven Proliferation

Evaluate proliferation after defined stimulation where the model supports it. Report viability, background proliferation, and the responding population so a change in total cell number is not mistaken for an antigen-specific response.

Orthogonal Functional Follow-Up

Where compatible target cells are available, discuss a separately scoped recognition or cytotoxicity assay. Target antigen expression and HLA compatibility are reviewed before interpreting a killing result as antigen-specific.

Choose the Right Stimulation Strategy

Study formatBest suited research questionImportant distinction
Direct ex vivo stimulationIs a response detectable in the submitted immune-cell sample without prior expansion?A negative result can reflect low frequency or limited sample quality; it does not prove absence of a relevant T cell repertoire.
In vitro stimulation or expansionCan antigen-responsive cells be enriched for exploratory analysis?Expansion changes cell composition and sensitivity. Expanded results are not reported as direct ex vivo frequencies.
Antigen presentation co-cultureCan a defined antigen-loaded APC system stimulate compatible responder cells? The APC, antigen format, and HLA or MHC background determine what the assay can demonstrate.
De novo priming feasibilityCan a proposed model investigate induction of a new response?Priming is not assumed to be supported by any generic PBMC assay. Availability of an appropriate configuration must be assessed first.

Peptide stimulation can bypass antigen uptake and processing. A peptide-positive response therefore does not establish that a protein, RNA construct, or tumor cell naturally presents the same epitope. Projects investigating this step can connect with DC maturation and antigen presentation assays or cancer antigenic epitope studies.

Information Needed for a Study Proposal

  • Antigen information: sequence or identity, peptide-pool design, solubility, vehicle, concentration, and relevant quality information.
  • Biological samples: PBMCs or selected T cells, fresh or cryopreserved status, sample history, donor or subject pairing, and available quantity.
  • Recognition context: known HLA restriction, responding clones or cell systems already available, and whether endogenous processing is part of the question.
  • Comparison plan: baseline versus follow-up samples, candidate versus reference antigen, and desired assay endpoints.

Cell input, number of conditions, and replicate requirements are agreed before shipping. For limited samples, we prioritize the primary question rather than splitting the material across an impractical panel. Sample handling can be coordinated with PBMC isolation and immune cell preparation services.

From Feasibility Review to Data Interpretation

Clarify the Decision

Define the primary response, comparison groups, and whether a discovery screen or a focused confirmation experiment is required.

Agree Cells, Antigens, and Controls

Review cell compatibility and sample quality. Set stimulation conditions, controls, and preliminary response criteria before analyzing the study samples.

Measure the Agreed Endpoints

Run the selected assay modules with sample and assay controls. Keep direct ex vivo and expanded conditions traceable throughout data processing.

Review Specificity and Variability

Assess background, replicate agreement, donor variation, and failed controls. Report inconclusive results explicitly and propose a follow-up only where it addresses an unresolved question.

Controls and Reporting Principles

Controls are selected for the assay rather than used as a generic checklist. They may include unstimulated and vehicle conditions, an irrelevant antigen, a compatible positive stimulation control, and an appropriate reference sample. Positive nonspecific stimulation confirms responsiveness under that condition; it does not validate antigen specificity.

For ELISpot, the report should distinguish raw spot counts, background-adjusted counts, cell input, and the prespecified interpretation rule. For flow-based assays, the gating approach, viability assessment, and relevant control strategy accompany the result. Biological variation and technical replication are presented separately.

Project Deliverables

Assay-specific resultsELISpot counts and normalized frequencies, selected activation or proliferation measurements, and comparative plots as applicable.
QC and interpretationControl performance, background handling, replicate variability, and results classified according to the agreed criteria.
Data handoverAn interpretive report and the raw-data formats specified in the proposal; no automatic claim of a validated clinical assay.

Frequently Asked Questions

Does a positive ELISpot result demonstrate tumor-cell killing?

No. It demonstrates cytokine secretion under the tested stimulation conditions. Cytotoxicity and recognition of naturally antigen-expressing tumor cells require additional, appropriately controlled experiments.

Can samples collected before and after vaccination be compared?

A paired design can be considered if processing, storage, and cell input support a meaningful comparison. Differences in sample quality or in vitro expansion must be addressed in the study plan.

Can you validate every predicted neoantigen?

Feasibility depends on HLA context, cell availability, and a suitable recognition system. A negative result is interpreted within those limits rather than used as a universal rejection of the candidate.

Research Context

Independent pancreatic cancer vaccine studies published in 2023 and 2025 illustrate the value of measuring antigen-specific function alongside broader analyses of vaccine-associated T cell populations. They support distinguishing response detection from longer-term persistence. These clinical publications provide scientific context and are not evidence of clinical efficacy or assay performance for this service.
