Description ANGPT2 is a protein that affects blood vessel formation and permeability. It promotes angiogenesis and is induced by hypoxia and inflammation. In lymphatic endothelium, it works as an agonist, but in blood endothelium, it acts as a competitive antagonist. TIE2 inhibition by ANGPT2 in blood vessels is associated with vascular leak, inflammation, and metastasis. A monoclonal antibody against ANGPT2 has been developed and can detect ANGPT2 protein in humans using ELISA and IHC.

Target Background

Function (From Uniprot) Binds to TEK/TIE2, competing for the ANGPT1 binding site, and modulating ANGPT1 signaling. Can induce tyrosine phosphorylation of TEK/TIE2 in the absence of ANGPT1. In the absence of angiogenic inducers, such as VEGF, ANGPT2-mediated loosening of cell-matrix contacts may induce endothelial cell apoptosis with consequent vascular regression. In concert with VEGF, it may facilitate endothelial cell migration and proliferation, thus serving as a permissive angiogenic signal.
