Iron Metabolism in TME

Iron is an indispensable element for all living organisms and the most abundant transition metal in the human body, and it is widely involved in important life metabolic processes. It plays a cofactor role in a variety of biochemical reactions such as DNA synthesis, electron transport, oxygen transport, mitochondrial respiration, host defense and cellular signal transduction, and may lead to the formation of reactive oxygen species (ROS), which adversely affects genomic stability and may induce malignant transformation. Thus, iron metabolism is one of the key factors determining the normal and malignant fate of cells. As with the metabolism of other essential metals, iron metabolism includes absorption, transport, storage, efflux, death and utilization. There are numerous studies showing that iron overload due to dysregulation of intracellular iron metabolism is associated with tumorigenesis, proliferation and metastasis, and the body's iron stores were found to be positively associated with the risk of certain tumors, such as rectal, liver, kidney, lung and gastric cancers. In addition to the direct supply of iron and iron-related proteins, macrophages and neutrophils in the tumor microenvironment (TME) may affect iron metabolism in tumor cells by releasing cytokines, chemokines and growth factors that act on tumor cells and induce changes in the signals regulating iron metabolism.


The present study shows that tumor cells exhibit a stronger dependence on iron, while consuming it more readily. Despite numerous studies, the specific link between iron metabolism and the tumor microenvironment (TME) is still not fully explained. Alfa Cytology has established an innovative technology platform for the tumor microenvironment center and is developing a variety of technologies to provide high quality solutions and services to our global collaborators. We design scientific experiments to help provide new insights into abnormal iron metabolism, gain insights into the relationship between tumor microenvironment (TME) and iron metabolism, and the mechanism of abnormal iron metabolism and tumorigenesis and metastasis, with the aim of providing new methods for predicting prognosis and treatment of tumors.

Key technologies

Single-omics is relatively one-sided in the resolution of biological problems. By combining proteome and metabolome, Alfa Cytology performs statistical analysis such as normalization, comparative analysis and correlation analysis on bulk data from two levels of biomolecules in the tumor microenvironment, including proteome and metabolome, to establish the relationship between data from different levels of molecules. At the same time, we combined GO functional analysis, metabolic pathway enrichment, molecular interactions and other biofunctional analyses to systematically and comprehensively analyze the functions and regulatory mechanisms of iron transport proteins and iron metabolism-related biomolecules. The integrated analysis of the two molecular levels of histological data can verify each other on the one hand, and help to complement each other on the other hand, so as to finally achieve a comprehensive understanding of the general trend and direction of biological changes, propose a model of the mechanism of iron metabolism changes in the sub-tumor microenvironment, and screen out key metabolic pathways or proteins and metabolites for subsequent in-depth experimental analysis and application.
